Description : Intacef Tazo ( Ceftriaxone & Tazobactam ) – Intacef Tazo

Intacef tazo is a combination of synergistic Ceftriaxone Sodium and  Tazobactam Sodium available as a dry powder for reconstitution. Ceftriaxone Sodium is a third generation , semi-synthetic , broad spectrum, bactericidal cephalosporin with molecular weight of 661.61. It is a white to yellowish orange crystalline powder, which is rapidly soluble in water and sparingly soluble in alcohol. Tazobactam sodium is the derivative of triazoymethyl penicillanic acid sulfone. It is a white off – white dry powder soluble in aqueous solvent.

Pharmacology of Intacef Tazo 562.50 mg – Intacef Tazo

Firstly we must have to understand about the mechanism of action of Intacef tazo 562.50 mg. Ceftriaxone found in this injection is a beta – lactam antibiotic that produces its bacterial effect by inhibition of the cell wall synthesis. Ceftriaxone has high degree of stability in the presence of the beta – lactamases both penicillinase and cephalosporinases of the gram negative and gram positive bacteria. Ceftriaxone is a type of broad spectrum anti-biotic.

Tazobactam :

Tazobactam is semi – synthetic beta-lactamase inhibitor. Tazobactam exhibits a moderate antibacterial activity that is relative to its affinity for the penicillin binding proteins of the various bacterial strains. It is a potent, highly specific irreversible inhibitor of beta – lactamases and extended spectrum beta – lactamase produced by the gram negative and gram positive aerobes and aerobes. Tazobactams binds itself with these enzymes and blocks their destructive hydrolytic activity.

Addition of Tazobactams with Ceftriaxone provides the following additional benefits :

Enhanced efficacy against beta – lactamases and extended spectrum beta-lactamases producing organisms.

Synergistic activity ( Reduction in minimum inhibitory concentration for the combination versus those of each component ).

Pharmacokinetics of Intacef Tazo

Ceftriaxone is widely distributed through the most body fluids and the tissues including kidney, lungs, bone, soft tissues, biliary tracts and extravascular spaces, pleural fluids, peritoneal fluids, synovial fluids and pericardial fluids. Ceftriaxone has excellent antibacterial activity against a wide range of gram negative and gram positive organisms. Ceftriaxone because of its extensive protein binding and half life of 8 hours offers once daily dose potential. Ceftriaxone is deacetylated primarily in the liver and kidney, the renal elimination is by renal tubular and glomerular filtration. The combination of Ceftriaxone and Tazobactam offers a broad spectrum bactericidal activity along with inhibition of beta lactamases and extended spectrum beta – lactamase which are a major threat to clinical therapeutics today.

Gram negative aerobes are :

Actinobacillus spp., Borrelia burgdorferi, Citrobacter spp., Enterobacter aerogenes, Enterobacter cloacae, Escherichia coli, Hemophilus spp., klebsiella pneumoniae, klebsiella oxytoca, Moraxella spp., Neisseria spp., Pasterurella haemolytica, Pasterurella multocida, Pasterurella multocida, Proteus mirabilis, Proteusvulgaris, Pseudomonas aeruginosa, Salmonella spp., Serratia marcescens, Shigella spp.

Gram positive aerobes are :

Staphylococcus aureus, Staphylococcus epidermis, Staphylococcus hemolyticus, Streptococcus pyogenes, Streptococcus pneumoniae, Streptococcus viridans, Streptococcus agalactiae, Streptococcus epidermis, Bacillus spp.

Anaerobes :

Clostridium spp., Bacteroids fragilis and Fusobacterium spp.
